Biological Background: CLIC4 Function and Localization
- CLIC4, also known as p64H1 or mtCLIC, is a member of the chloride intracellular channel (CLIC) family, existing in both soluble and integral membrane forms.
- In its soluble state, CLIC4 functions as a glutaredoxin-like oxidoreductase, catalyzing thiol-disulfide exchange reactions using reduced glutathione as an electron donor. Related references: PMID:25581026 PMID:37759794
- Under oxidizing conditions, CLIC4 can insert into membranes to form voltage-dependent, multi-ion conductive channels; membrane insertion is redox-regulated. Related references: PMID:16176272
- CLIC4 plays roles in angiogenesis, endothelial cell proliferation, and maintenance of apical-basolateral polarity during mitosis and cytokinesis.
- The protein localizes to multiple subcellular compartments, including the cytoplasm, cytoskeleton, centrosomes, cytoplasmic vesicles, nucleus, cell membrane, mitochondria, cell junctions, and endoplasmic reticulum.
- Post-translational modifications include acetylation and phosphorylation, as identified by proteomics.

- The immunogen corresponds to the N-terminal region (aa 1–100) of human CLIC4. This region may be effective for detecting both soluble and membrane-inserted forms; validate specificity in your experimental system.
- For Western blot, expect a band at approximately 29 kDa under reducing conditions. Use appropriate positive controls (e.g., HeLa or HEK293 lysates) and confirm with known expression patterns.
- In ELISA, this antibody can be used for direct antigen detection or as a capture/detection reagent. Consider compatibility with your assay format and sample matrix.
- Reported cross-reactivity with mouse and rat CLIC4; verify sequence homology for your application.
