Biological Background: CLEC7A Function and Localization
- CLEC7A (Dectin-1) is a member of the C-type lectin family, also known as beta-glucan receptor, CLECSF12, and DC-associated C-type lectin 1.
- It functions as a pattern recognition receptor (PRR) that specifically recognizes beta-1,3- and beta-1,6-linked glucans from fungal and bacterial cell walls. Related references: PMID:11567029 PMID:12423684
- Upon ligand binding, CLEC7A recruits SYK via its ITAM motif, leading to activation of NF-kappa-B and MAP kinase pathways, which induce pro-inflammatory cytokines and chemokines.
- It mediates phagocytosis of Candida albicans conidia and enhances cytokine production in macrophages and dendritic cells. Related references: PMID:17230442
- CLEC7A is also involved in T-cell activation, binding to T-cells independently of surface glycans and stimulating their proliferation.
- Subcellularly, the protein localizes to the cell membrane and cytoplasm. It is highly expressed in peripheral blood leukocytes, dendritic cells, spleen, bone marrow, lung, muscle, stomach, and placenta.
- Key structural features include disulfide bonds, glycosylation, and a transmembrane domain, reflecting its role as a signaling receptor in innate immunity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a peptide within amino acids 140–240 of human CLEC7A (NP_922938.1); verify epitope accessibility under your experimental conditions.
- For Western blot, anticipate a band near 28 kDa (predicted MW ~28 kDa; UniProt: 27.6 kDa). Run appropriate positive controls (e.g., leukocyte or dendritic cell lysates).
- This antibody can be used in ELISA; consider optimizing coating antigen and antibody dilution through checkerboard titrations.
- Note that cross-reactivity with mouse and rat has been reported; confirm specificity in your model system.
