Biological Background: CLEC5A Function and Localization
-
CLEC5A, also known as myeloid DAP12-associating lectin 1 (MDL-1) or CLECSF5, is a type II transmembrane protein belonging to the C-type lectin domain family.
-
Functions as a cell surface receptor that signals via TYROBP, acting as a positive regulator of osteoclastogenesis and inflammatory responses.

Serves as a critical macrophage receptor for dengue virus serotypes 1-4. Binding of the virus triggers TYROBP phosphorylation and pro-inflammatory cytokine release, but does not mediate viral entry.
Related references: PMID:18496526 PMID:21566123 -
Localized to the cell membrane.
-
Highly expressed in bone marrow, with lower levels in synovium, lung, and bronchus. Expressed in peripheral blood monocytes and monocyte/macrophage cell lines, but down-regulated upon monocyte differentiation into dendritic cells.
Related references: PMID:20212065 PMID:10449773 -
Contains a single transmembrane domain and a C-type lectin domain; classified as a host cell receptor for virus entry and involved in innate immunity.
Experimental Guidance and Technical Tips
-
The immunogen corresponds to the extracellular domain (amino acids 29-188) of human CLEC5A, suggesting recognition of the native protein and applicability for detecting both endogenous and recombinant CLEC5A.
-
For Western blot, optimize blocking and antibody dilution; the predicted molecular weight of CLEC5A is approximately 22 kDa, so use appropriate gel acrylamide percentage and transfer settings for low-molecular-weight proteins.
-
The antibody is reported to cross-react with human, mouse, and rat CLEC5A, making it suitable for cross-species studies; however, validate specificity in your specific model system using appropriate controls.
-
For ELISA, this polyclonal antibody may be used as a capture or detection reagent; consider pairing with a monoclonal antibody for enhanced specificity, and optimize assay conditions.
