Biological Background: CLEC4E (Mincle) Function and Localization

  • CLEC4E, also known as C-type lectin domain family 4 member E, macrophage-inducible C-type lectin (Mincle), or C-type lectin superfamily member 9, is a calcium-dependent lectin and pattern recognition receptor (PRR) of the innate immune system. Related references: PMID:18490740, PMID:18509109
  • It recognizes damage-associated molecular patterns (DAMPs) from abnormal self and pathogen-associated molecular patterns (PAMPs) from bacteria and fungi, including mycobacterial trehalose 6,6'-dimycolate (TDM) and fungal alpha-mannose residues. Related references: PMID:18776906, PMID:20008526, PMID:23602766, PMID:19171887
  • Upon ligand binding, CLEC4E associates with the Fc receptor gamma chain (FCER1G) to form a signaling complex, leading to ITAM phosphorylation and activation of SYK, CARD9, and NF-kappa-B pathways, thereby promoting maturation of antigen-presenting cells and Th1/Th17 polarization. Related references: PMID:23602766
  • CLEC4E localizes to the cell membrane, cell projections, and the phagocytic cup, consistent with its role in immune surveillance and phagocytosis.
  • It is highly expressed in macrophages and dendritic cells upon stimulation with bacterial glycolipids or pro-inflammatory cytokines, linking innate sensing to adaptive immunity. Related references: PMID:10528209, PMID:23602766
  • The protein contains structural features typical of C-type lectins, including a transmembrane helix, a signal-anchor, disulfide bonds, and glycosylation sites, and it binds calcium and other metals essential for carbohydrate recognition.
  • Gene family: CLEC4E belongs to the C-type lectin superfamily. Its mouse gene ID is 56619, and the human ortholog is 26253.

Experimental Guidance and Technical Tips

  • The immunogen is a recombinant fusion protein spanning amino acids 120–200 of mouse CLEC4E (NP_064332.1), corresponding to the predicted extracellular C-type lectin domain. This region is expected to be exposed on the cell surface and accessible for antibody binding.
  • For Western blot (WB), the predicted molecular weight is ~24 kDa; consider running appropriate positive controls such as stimulated macrophage lysates.
  • In ELISA, the antibody may capture and detect recombinant or native CLEC4E; validate assay conditions using established antigen preparations.
  • The antibody shows cross-reactivity with rat, so it may be suitable for comparative studies. Optimize dilutions for each species and application.
  • As a polyclonal antibody, it may recognize multiple epitopes, providing robust signal but potential batch-to-batch variation; consider lot-specific validation in critical experiments.
