  • Also known as Dectin-3, CD368, and CLECSF8, CLEC4D is a member of the C-type lectin family that functions as a pattern recognition receptor (PRR) in the innate immune system.
  • CLEC4D recognizes damage-associated molecular patterns (DAMPs) and pathogen-associated molecular patterns (PAMPs) from bacteria and fungi, including α-mannans on Candida albicans hyphae and mycobacterial trehalose 6,6'-dimycolate (TDM). Related references: PMID:23602766 PMID:23911656
  • Upon ligand engagement, CLEC4D likely forms a complex with Fc receptor gamma chain (FCER1G) via CLEC4E (Mincle), leading to phosphorylation of the ITAM motif, SYK-CARD9-NF-kB activation, and subsequent maturation of antigen-presenting cells with induction of T-helper 1 and 17 responses.   • For Western blotting, the predicted molecular weight is ~25 kDa, but glycosylated forms may migrate higher. Use appropriate positive controls such as THP-1 cells stimulated with cytokines (e.g., TNF or IFN-gamma) to enhance CLEC4D expression.
