Biological Background: CLEC2D Function and Localization

  • CLEC2D (C-type lectin domain family 2 member D), also known as LLT1, OCIL, or CLAX, is a transmembrane receptor belonging to the C-type lectin superfamily. It is encoded by the CLEC2D gene and exists in multiple isoforms due to alternative splicing.
  • Acts as a receptor for KLRB1 (CD161) on natural killer (NK) cells, inhibiting NK cell-mediated cytotoxicity and protecting target cells from lysis.
    Related references: PMID:16339513, PMID:20843815
  • Functions as a negative regulator of bone remodeling by inhibiting osteoclast formation and bone resorption.
    Related references: PMID:14753741, PMID:15123656
  • Modulates immune responses by regulating the release of interferon-gamma, linking innate immunity to bone homeostasis.
    Related references: PMID:15104121
  • Binds to high molecular weight sulfated glycosaminoglycans, which may mediate its interactions with extracellular matrix components.
    Related references: PMID:15123656
  • Localizes predominantly to the cell membrane, but also detected in the endoplasmic reticulum, consistent with its role as a transmembrane signaling receptor.
  • Expressed in peripheral blood leukocytes, lymph node, thymus, and spleen, with isoforms present in T cells, B cells, and at lower levels in NK cells and dendritic cells.
  • Features a typical C-type lectin-like domain, signal-anchor transmembrane topology, disulfide bonds, and N-linked glycosylation, as indicated by sequence analysis and structural data (UniProt keywords).

Experimental Guidance and Technical Tips

  • The immunogen comprises amino acids 60–154 of human CLEC2D (NP_037401.1), corresponding to the extracellular C-type lectin-like domain; the antibody is expected to recognize native, surface-exposed CLEC2D. For applications requiring detection of endogenous protein, consider validation in relevant cell lines (e.g., NK cells, B cells, osteoblasts) or primary cell populations.
  • For Western blot, the predicted molecular weight is ~22 kDa based on the amino acid sequence; however, glycosylation may result in a higher apparent molecular mass. Include appropriate positive controls and molecular weight markers. The antibody may also detect isoforms arising from alternative splicing.
  • For ELISA, the antibody can be used as a capture or detection reagent; the recombinant immunogen (aa 60–154) can serve as a standard. Cross-reactivity with mouse CLEC2D has been reported, but sequence homology should be verified if used in mouse models.
  • If used in immunofluorescence or flow cytometry, permeabilization may be necessary to access intracellular ER-localized pools, while surface staining can be performed on live, non-permeabilized cells.
  • Always optimize antibody dilution and incubation conditions empirically for each application and sample type. This antibody is supplied as a polyclonal serum and may exhibit lot-to-lot variability; consistency should be verified in critical assays.
