Biological Background: CLEC2D Function and Localization
- CLEC2D (C-type lectin domain family 2 member D), also known as LLT1, OCIL, or CLAX, is a transmembrane receptor belonging to the C-type lectin superfamily. It is encoded by the CLEC2D gene and exists in multiple isoforms due to alternative splicing.
- Acts as a receptor for KLRB1 (CD161) on natural killer (NK) cells, inhibiting NK cell-mediated cytotoxicity and protecting target cells from lysis.

- The immunogen comprises amino acids 60–154 of human CLEC2D (NP_037401.1), corresponding to the extracellular C-type lectin-like domain; the antibody is expected to recognize native, surface-exposed CLEC2D. For applications requiring detection of endogenous protein, consider validation in relevant cell lines (e.g., NK cells, B cells, osteoblasts) or primary cell populations.
- For Western blot, the predicted molecular weight is ~22 kDa based on the amino acid sequence; however, glycosylation may result in a higher apparent molecular mass. Include appropriate positive controls and molecular weight markers. The antibody may also detect isoforms arising from alternative splicing.
- For ELISA, the antibody can be used as a capture or detection reagent; the recombinant immunogen (aa 60–154) can serve as a standard. Cross-reactivity with mouse CLEC2D has been reported, but sequence homology should be verified if used in mouse models.
- If used in immunofluorescence or flow cytometry, permeabilization may be necessary to access intracellular ER-localized pools, while surface staining can be performed on live, non-permeabilized cells.
