Biological Background: CLCC1 Function and Localization
- CLCC1, also known as MCLC, RP32, ER anion channel 1 (ERAC1), and Mid-1-related chloride channel protein, is a chloride channel CLIC-like protein encoded by the CLCC1 gene.
- It functions as an anion-selective channel with Ca(2+)-dependent and voltage-independent gating, permeable to small monovalent anions with a selectivity of bromide > chloride > nitrate > fluoride.
- Localized to the endoplasmic reticulum (ER) membrane, it mediates chloride efflux to compensate for the loss of positive charges from the ER lumen upon Ca(2+) release.
- Contributes to the maintenance of ER Ca(2+) pools and activation of the unfolded protein response, preventing accumulation of misfolded proteins, which is critical for motor neuron and retinal photoreceptor survival. Related references: PMID:25698737 PMID:30157172 PMID:37142673
- In the retina, CLCC1 is extensively expressed in the lamina cribrosa, optic nerve, ganglion cell layer, inner nuclear layer, outer nuclear layer, and retinal pigment epithelium.
- Post-translational modifications include phosphorylation, and the protein undergoes alternative splicing. It is classified under keywords such as chloride channel, ion transport, endoplasmic reticulum, and reference proteome.
Experimental Guidance and Technical Tips
- The immunogen used to generate this antibody corresponds to amino acids 302-501 of human CLCC1, targeting the C-terminal region; consider this when validating binding to potential isoforms or splice variants.
- Recommended applications include Western blot (WB), immunohistochemistry (paraffin-embedded, IHC-P), and ELISA. Optimize antibody dilution and experimental conditions using appropriate positive and negative controls for your sample type.
- This antibody cross-reacts with both human and mouse CLCC1, making it suitable for studies in mouse models; however, perform validation in the specific tissue or cell line of interest.
- For WB, the predicted molecular weight of CLCC1 is 62 kDa; confirm band specificity with blocking peptides or siRNA knockdown as needed.
