Biological Background: CLCA1 Function and Localization
- CLCA1, also known as calcium-activated chloride channel regulator 1 or CaCC-1, belongs to the calcium-activated chloride channel family.
- It is proposed to mediate calcium-activated chloride conductance and plays critical roles in goblet cell metaplasia, mucus hypersecretion, and airway hyperresponsiveness (AHR).
- Involved in the regulation of mucus production and secretion by goblet cells, and induces MUC5AC expression.
- Plays a role in tissue inflammation as part of the innate immune response and may function as a tumor suppressor.
- Highly expressed in the small intestine, colon (especially intestinal basal crypt epithelia and goblet cells), appendix, and bronchial epithelium; weakly expressed in uterus, testis, and kidney.
- Subcellularly localized to the cell membrane and secreted into the extracellular space.
- Features autocatalytic cleavage, glycosylation, and metal-binding domains (zinc), characteristic of metalloproteases.
- The protein has a molecular weight of approximately 100 kDa (100226 Da).

- The immunogen spans amino acids 22–150 in the N-terminal region; consider that this antibody may detect full-length and cleaved fragments.
- For Western blotting, CLCA1 migrates at ~100 kDa; validate band specificity using relevant tissue lysates (e.g., colon, small intestine).
- In IF-P, anticipate membrane and possibly secreted localization; include appropriate controls to confirm staining.
