Biological Background: CLCA1 Function and Localization

  • CLCA1, also known as calcium-activated chloride channel regulator 1 or CaCC-1, belongs to the calcium-activated chloride channel family.
  • It is proposed to mediate calcium-activated chloride conductance and plays critical roles in goblet cell metaplasia, mucus hypersecretion, and airway hyperresponsiveness (AHR).
  • Involved in the regulation of mucus production and secretion by goblet cells, and induces MUC5AC expression.
  • Plays a role in tissue inflammation as part of the innate immune response and may function as a tumor suppressor.
  • Highly expressed in the small intestine, colon (especially intestinal basal crypt epithelia and goblet cells), appendix, and bronchial epithelium; weakly expressed in uterus, testis, and kidney.
  • Subcellularly localized to the cell membrane and secreted into the extracellular space.
  • Features autocatalytic cleavage, glycosylation, and metal-binding domains (zinc), characteristic of metalloproteases.
  • The protein has a molecular weight of approximately 100 kDa (100226 Da).
