Biological Background: Clathrin heavy chain 1 Function and Localization
- Clathrin heavy chain 1 (CLTC, also known as CLH-17) is the major structural component of the polyhedral coat of coated pits and vesicles, essential for receptor-mediated endocytosis and intracellular trafficking.
- Acts as a component of the TACC3/ch-TOG/clathrin complex, contributing to kinetochore fiber stabilization by forming inter-microtubule bridges, and is required for maintenance of kinetochore fiber tension (PubMed:15858577, PubMed:16968737, PubMed:21297582, PubMed:23532825). Related references: PMID:15858577 PMID:16968737 PMID:21297582 PMID:23532825
- Involved in early autophagosome formation (PubMed:20639872). Related references: PMID:20639872
- Interaction with DNAJC6 mediates the recruitment of HSPA8 to the clathrin lattice, creating local destabilization that promotes uncoating (By similarity).
- Subcellular localization includes cytoplasmic vesicle membranes, coated pits, melanosomes, and the mitotic spindle, reflecting its diverse roles in membrane traffic and cell division.
- Post-translational modifications include acetylation and phosphorylation, and it is implicated in processes such as cell cycle progression, mitosis, and autophagy.
- Alternative splicing generates multiple isoforms; the canonical isoform has a molecular weight of approximately 192 kDa (191,615 Da per UniProt).
Experimental Guidance and Technical Tips
- The immunogen (aa 1451-1675) resides within a conserved C-terminal region, supporting applications such as western blot (WB), immunofluorescence/immunocytochemistry (IF/ICC), and ELISA; optimizing dilution factors for each application is recommended.
- Due to high sequence identity across species, cross-reactivity with mouse and rat is expected; if using non-human samples, validate in the relevant sample system.
- For WB, note the large molecular weight (~192 kDa); use appropriate gel percentages (e.g., 6–8% SDS-PAGE) and extended transfer times for efficient blotting.
- As a rabbit polyclonal antibody, it may recognize multiple epitopes, enhancing signal but requiring careful specificity controls such as knockout validation.
