Biological Background: Clathrin Heavy Chain Function and Localization
- Clathrin heavy chain 1 (CLTC) is the major structural component of the polyhedral coat of clathrin-coated pits and vesicles, mediating both plasma membrane and trans-Golgi network trafficking.
- Acts as a component of the TACC3/ch-TOG/clathrin complex that stabilizes kinetochore fibers of the mitotic spindle by bridging microtubules, essential for maintaining kinetochore fiber tension during mitosis. Related references: PMID:15858577 PMID:16968737 PMID:21297582 PMID:23532825
- Plays a role in early autophagosome formation. Related references: PMID:20639872
- Interaction with DNAJC6 recruits HSPA8 to the clathrin lattice, locally destabilizing it to promote uncoating (By similarity).

- Subject to post-translational modifications including acetylation and phosphorylation, with alternative splicing generating multiple isoforms.
