Biological Background: Clathrin heavy chain 1 Function and Localization
- Clathrin heavy chain 1 (CLTC) is a major structural protein of coated pits and vesicles, also known as clathrin heavy chain on chromosome 17 (CLH-17).
- Clathrin forms the polyhedral coat of coated pits and vesicles, mediating endocytosis and intracellular trafficking through interactions with adapter protein complexes at the plasma membrane and trans-Golgi network.
- Acts as a component of the TACC3/ch-TOG/clathrin complex, which contributes to stabilization of kinetochore fibers of the mitotic spindle by acting as an inter-microtubule bridge and is required for maintenance of kinetochore fiber tension. - Plays a role in early autophagosome formation. Related reference: PMID:20639872
- Interaction with DNAJC6 is thought to mediate recruitment of HSPA8 to the clathrin lattice, creating local destabilization that promotes uncoating of clathrin cages.
- Subcellularly localized to cytoplasmic vesicle membranes, coated pits, melanosomes, the cytoplasm, cytoskeleton, and mitotic spindle.
- Subject to acetylation and phosphorylation, with alternative splicing producing multiple isoforms; mutations are associated with intellectual disability.
