Biological Background: Clathrin heavy chain 1 Function and Localization

  • Clathrin heavy chain 1 (CLTC) is a major structural protein of coated pits and vesicles, also known as clathrin heavy chain on chromosome 17 (CLH-17).
  • Clathrin forms the polyhedral coat of coated pits and vesicles, mediating endocytosis and intracellular trafficking through interactions with adapter protein complexes at the plasma membrane and trans-Golgi network.
  • Acts as a component of the TACC3/ch-TOG/clathrin complex, which contributes to stabilization of kinetochore fibers of the mitotic spindle by acting as an inter-microtubule bridge and is required for maintenance of kinetochore fiber tension. Related references: PMID:15858577 PMID:16968737 PMID:21297582 PMID:23532825
  • Plays a role in early autophagosome formation. Related reference: PMID:20639872
  • Interaction with DNAJC6 is thought to mediate recruitment of HSPA8 to the clathrin lattice, creating local destabilization that promotes uncoating of clathrin cages.
  • Subcellularly localized to cytoplasmic vesicle membranes, coated pits, melanosomes, the cytoplasm, cytoskeleton, and mitotic spindle.
  • Subject to acetylation and phosphorylation, with alternative splicing producing multiple isoforms; mutations are associated with intellectual disability.

Experimental Guidance and Technical Tips

  • The immunogen used is a synthetic peptide from aa 1550-1650 of human Clathrin heavy chain; consider that this region is conserved across human, mouse, and rat, supporting the observed cross-reactivity.
  • For Western blot (WB), a band around 192kDa is expected; consider optimizing lysate preparation and gel conditions for large proteins.
  • For ELISA, validate the antibody with the relevant recombinant protein or peptide to confirm specificity.
  • As a polyclonal antibody, it may recognize multiple clathrin isoforms; confirm specificity in your system using appropriate controls.
