Biological Background: CISD2 Function and Localization
- CISD2 (CDGSH iron-sulfur domain-containing protein 2), also known as ERIS, Miner1, and NAF-1, is a 135 amino acid protein with a predicted molecular weight of approximately 15 kDa.
- It contains a CDGSH domain that coordinates a 2Fe-2S cluster, characteristic of the mitoNEET protein family.
- CISD2 is a key regulator of autophagy, functioning at the endoplasmic reticulum (ER) by mediating the interaction between BCL2 and BECN1, which is required for BCL2-mediated depression of ER Ca²⁺ stores during autophagy.
- It participates in BIK-initiated autophagy without affecting BIK-dependent caspase activation, indicating a specific role in autophagic rather than apoptotic pathways.
- The protein localizes to both the ER membrane and the mitochondrial outer membrane, linking these organelles in cellular homeostasis.
- CISD2 is implicated in life span control and its dysfunction is associated with Wolfram syndrome 2 (WFS2), a disorder featuring diabetes mellitus and deafness.
- It is broadly expressed in tissues including testis, small intestine, kidney, lung, brain, heart, pancreas, and platelets.
Experimental Guidance and Technical Tips
- For Western blot, expect a band at approximately 15 kDa; use a positive control lysate from human, mouse, or rat cell lines or tissues known to express CISD2 (e.g., testis, kidney, or brain).
- In immunohistochemistry (IHC-P), optimize antigen retrieval (e.g., heat-induced epitope retrieval in citrate buffer pH6) and validate using tissue sections with abundant expression, such as pancreas or small intestine.
- For ELISA, consider pairing with a CISD2-specific capture antibody if designing a sandwich assay; perform a checkerboard titration to determine optimal concentrations.
- Since the immunogen covers amino acids 61–135, the antibody may recognize both endogenous and recombinantly expressed CISD2; cross-reactivity with mouse and rat homologs has been demonstrated, but additional validation is recommended for other species.
- Include appropriate positive and negative controls (e.g., CISD2 knockdown lysates or tissues) to confirm specificity in your experimental system.
