Biological Background: CIRBP Function and Localization

  • CIRBP (Cold-inducible RNA-binding protein), also known as A18 hnRNP and glycine-rich RNA-binding protein CIRP, is encoded by the CIRBP gene in humans.
  • Acts as a cold-inducible mRNA binding protein, playing a protective role in the genotoxic stress response by stabilizing transcripts of genes involved in cell survival.
  • Functions as both a translational activator and repressor; it binds specifically to the 3'-UTRs of stress-responsive transcripts such as RPA2 and TXN.
  • When overexpressed, promotes the assembly of stress granules (SGs), which are cytoplasmic aggregates of stalled translation preinitiation complexes.
  • Localizes primarily to the nucleus and nucleoplasm, and can shuttle to the cytoplasm under stress conditions.
  • Ubiquitously expressed across tissues, consistent with its role in cellular stress responses.
  • Post-translational modifications include phosphorylation (as indicated by the keyword "Phosphoprotein").
  • The protein has alternative splice variants, contributing to functional diversity.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the full-length human CIRBP protein (aa 1-172), which is ideal for detecting total CIRBP in samples.
  • For Western blotting (WB), the predicted molecular weight is approximately 19 kDa; however, post-translational modifications might alter migration.
  • Immunoprecipitation (IP) and ELISA applications may benefit from the high affinity and specificity of this rabbit monoclonal antibody.
  • For immunofluorescence (IF) and immunohistochemistry (IHC), consider optimizing fixation and permeabilization protocols, as CIRBP can shuttle between nucleus and cytoplasm.
