Biological Background: Cip4 Function and Localization
- Cip4, also known as Cdc42-interacting protein 4, is encoded by the TRIP10 gene and also referred to as STP, HSTP, STOT, and TRIP-10.
- Insulin signaling: Required for translocation of glucose transporter GLUT4 to the plasma membrane in response to insulin (by similarity).
- Endocytic membrane tubulation: Coordinates membrane tubulation with reorganization of the actin cytoskeleton during endocytosis. Binds to lipids such as phosphatidylinositol 4,5-bisphosphate and phosphatidylserine to promote membrane invagination and tubule formation.
- Actin polymerization: Promotes CDC42-induced actin polymerization by recruiting WASL/N-WASP, which activates the Arp2/3 complex; this may aid in fission of membrane tubules to form endocytic vesicles.
- Podosome formation: Required for the formation of podosomes, actin-rich adhesion structures in monocyte-derived cells, and may be involved in lysosomal retention of FASLG/FASL.
- Subcellular localization: Found in the cytoplasm, cytoskeleton, cell cortex, lysosome, Golgi apparatus, cell membrane, phagocytic cup, and perinuclear region.
- Tissue distribution: Widely expressed in brain, colon, heart, kidney, liver, lung, megakaryocyte, ovary, pancreas, peripheral blood lymphocytes, placenta, prostate, skeletal muscle, small intestine, spleen, testis, thymus, and trachea.
- Structural features: Contains an SH3 domain and a coiled coil region; is a phosphoprotein involved in lipid binding and endocytosis.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to the N-terminal region (amino acids 1-100) of human Cip4. This antibody is expected to recognize the full-length protein and potentially N-terminal isoforms.
- Predicted molecular weight is 68 kDa; confirm by Western blot with appropriate positive controls (e.g., cell lines expressing Cip4) and use a reliable protein ladder.
- Validated applications: Western blot (WB) and ELISA. For WB, consider optimizing antibody dilution (e.g., 1:500-1:2000) and blocking conditions in your specific experimental system. For ELISA, test with recombinant protein or peptide to establish standard curves.
- Cross-reactivity is observed with human and mouse Cip4, indicating sequence conservation in the immunogen region. Validate reactivity in your target species and sample type.
- When using for endocytosis or cytoskeletal studies, consider appropriate stimulation conditions (e.g., insulin treatment for GLUT4 translocation assays).
- Store at -20°C or as recommended; avoid repeated freeze-thaw cycles to maintain stability.
