Biological Background: CIB1 Function and Localization

• Calcium and integrin-binding protein 1 (CIB1, also known as calmyrin, KIP1, SIP2-28) is encoded by the CIB1 gene. Multiple transcript variants exist due to alternative splicing.
• Functions as a calcium-binding regulatory protein involved in cell adhesion and integrin signaling, negatively regulating alpha-IIb/beta3 activation in thrombin-stimulated megakaryocytes to prevent platelet aggregation.
• Positively regulates cell migration on fibronectin in a CDC42-dependent manner, and promotes PTK2/FAK1 recruitment to focal adhesions, playing a key role in focal adhesion formation.
