Biological Background: cIAP2 Function and Localization

  • Baculoviral IAP repeat-containing protein 3 (BIRC3), also known as cIAP2, API2, and hIAP1, is a member of the inhibitor of apoptosis (IAP) family characterized by the presence of one or more baculovirus IAP repeat (BIR) domains and a RING finger domain.
  • Functions as a multi-functional regulator of apoptosis, inflammatory signaling, and immunity by acting as an E3 ubiquitin-protein ligase, mediating ubiquitination of key signaling molecules such as RIPK1, RIPK2, CASP3, CASP7, CASP8, IKBKE, TRAF1, and BCL10.
  • Positively regulates canonical NF-kB signaling while suppressing constitutive activation of the non-canonical pathway, thereby balancing inflammatory and cell survival responses.
  • Plays a critical role in innate immunity by regulating pattern recognition receptors (PRRs), including Toll-like receptors (TLRs), NOD-like receptors (NLRs), and RIG-I-like receptors (RLRs), influencing downstream cytokine production.

Experimental Guidance and Technical Tips

  • The immunogen was designed against amino acids 100–130 of human cIAP2 (NP_001156.1), a region within the BIR domains; the resulting polyclonal antibody is expected to recognize the full-length protein (~68 kDa) in Western blotting.
  • For Western blot (WB), use 5% nonfat milk or BSA for blocking and overnight primary incubation at 4°C; a band at approximately 68 kDa should be detected. Consider using lysates from cells with known cIAP2 expression (e.g., lymphoid lines) as positive controls.
  • For immunohistochemistry (IHC-P), antigen retrieval may be required; validate staining specificity with tissue sections from spleen or thymus and compare with normal IgG controls.
  • In immunofluorescence (IF/ICC), observe cytoplasmic and nuclear localization; co-staining with organelle markers can confirm subcellular distribution.
