Biological Background: cIAP1 Function and Localization
- The target protein is Baculoviral IAP repeat-containing protein 2 (BIRC2), also known as cIAP1, HIAP2, and RNF48. Encoded by the BIRC2 gene in humans, it belongs to the inhibitor of apoptosis (IAP) family.
- cIAP1 is a multifunctional E3 ubiquitin-protein ligase that regulates both canonical and non-canonical NF-κB signaling, acting as a positive regulator of the canonical pathway and suppressing constitutive activation of the non-canonical pathway. It also modulates apoptosis by targeting caspases (CASP3, CASP7, CASP8) and RIP kinases for ubiquitination.
- The protein targets numerous substrates, including RIPK1, RIPK2, TRAF2, and DIABLO/SMAC, and can also function in the NEDD8 conjugation pathway, neddylating effector caspases to inactivate them.
- cIAP1 plays a critical role in innate immune signaling by regulating pattern recognition receptors (PRRs) such as Toll-like receptors, Nod-like receptors, and RIG-I-like receptors.
- It protects cells from ripoptosome formation by ubiquitinating RIPK1 and CASP8, thereby preventing caspase-dependent and -independent cell death.
- Subcellular localization includes both cytoplasm and nucleus, consistent with its roles in signal transduction and transcriptional regulation.
- cIAP1 is widely expressed in fetal and adult tissues, with highest levels in skeletal muscle, thymus, testis, ovary, and pancreas, and low or absent in brain and peripheral blood leukocytes.
- Key structural features include three BIR (baculoviral IAP repeat) domains, a CARD domain, and a RING-type zinc finger at the C-terminus, which mediates its E3 ubiquitin ligase activity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 331-420 within the C-terminal region of human cIAP1, which includes part of the RING domain. Cross-reactivity with mouse and rat is reported, but validation in your specific experimental system is recommended.
- Recommended applications include Western blot (predicted band at ~70 kDa), immunohistochemistry (IHC-P), immunofluorescence (IF/ICC), and ELISA. For IHC-P and IF/ICC, optimize antigen retrieval and permeabilization conditions.
- As a polyclonal antibody, lot-to-lot consistency may vary; verify performance with each new batch.
