Biological Background: Chromogranin A Function and Localization
- Chromogranin A (CgA, also known as pituitary secretory protein I, SP-I) is a member of the granin family of neuroendocrine secretory proteins, encoded by the CHGA gene (Gene ID: 1113).
- It strongly inhibits glucose-induced insulin release from the pancreas, contributing to the regulation of glucose homeostasis.
- CgA acts as a non-competitive nicotinic cholinergic antagonist to inhibit catecholamine release from chromaffin cells and noradrenergic neurons (PubMed:15326220). Related references: PMID:15326220
- It displays broad-spectrum antibacterial activity against Gram-positive bacteria S. aureus and M. luteus, and Gram-negative bacteria E. coli and P. aeruginosa (PubMed:15723172, PubMed:24723458). Related references: PMID:15723172 PMID:24723458
- CgA can induce mast cell migration, degranulation and production of cytokines and chemokines, implicating it in innate immune responses (PubMed:21214543). Related references: PMID:21214543
- Chromogranin A may modulate cardiac function and blood pressure regulation (PubMed:18541522). Related references: PMID:18541522
- In endocrine cells, CgA regulates secretory granule biogenesis by up-regulating protease nexin 1 (SERPINE2) via a cAMP-PKA-SP1 pathway.
- Subcellularly, CgA localizes to secretory vesicles including neuronal dense core vesicles and is secreted; it is detected in brain, cerebrospinal fluid, and urine.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1–100) of human Chromogranin A, which is exposed in the mature secreted protein. Consider using a blocking peptide for specificity controls in immunohistochemistry or ELISA.
- For IHC-P, antigen retrieval in citrate buffer may be required; validate with known positive control tissues such as pancreas or adrenal gland.
- For IF-P, optimize fixation and permeabilization conditions for your sample type. Co-staining with a granule marker can confirm localization.
- In ELISA, this antibody may be used as a capture or detection reagent; pairing with a different epitope-specific antibody is recommended for sandwich assays.
- Cross-reactivity with mouse and rat has been reported, but verify performance in the relevant species by pilot experiments.
