Biological Background: CHRNA7 (Alpha-7 nAChR) Function and Localization
- CHRNA7 encodes the alpha-7 subunit of neuronal nicotinic acetylcholine receptors (nAChRs), and alternative splicing produces multiple isoforms; this immunogen is derived from isoform NP_001177384.1 (extracellular domain, aa 23-230).
- CHRNA7 assembles into homopentameric cation channels with high calcium permeability and fast desensitization kinetics, mediating excitatory neurotransmission in the central nervous system. Related references: PMID:31560909, PMID:33735609, PMID:38382524
- It can also form heteropentamers with CHRNB2, primarily in basal forebrain cholinergic neurons, and modulates neurotransmitter release including dopamine, glutamate, and GABA. Related references: PMID:33239400
- CHRNA7 is expressed in non-neuronal cells such as lymphocytes, monocytes, and macrophages, where it regulates inflammatory responses via the cholinergic anti-inflammatory pathway. Related references: PMID:12508119, PMID:16968406, PMID:25259522
- The receptor is localized at the postsynaptic cell membrane, consistent with its role in synaptic transmission.
- CHRNA7 is a glycoprotein containing disulfide bonds, which are critical for proper folding and receptor assembly.
- Predominantly expressed in neuronal cells, with high levels in brain regions such as the hippocampus and cortex, and also detected in macrophages at the protein level. Related references: PMID:8145738, PMID:12508119
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal extracellular domain (aa 23-230) of human CHRNA7, which is highly conserved across human, mouse, and rat, supporting the observed cross-reactivity.
- For Western blot, the predicted molecular weight is ~54 kDa; consider using appropriate protein extraction and blocking conditions to detect the glycosylated mature form.
- In immunohistochemistry and immunocytochemistry, membrane localization is expected; include appropriate controls and optimize antigen retrieval for paraffin-embedded tissues.
- ELISA can be used for quantitative detection; ensure coating conditions are suitable for the recombinant immunogen or native protein.
