Biological Background: CHRAC1 Function and Localization

  • CHRAC1, also known as chromatin accessibility complex 15 kDa protein (CHRAC-15) and DNA polymerase epsilon subunit p15, is a small protein of approximately 14.7 kDa encoded by the CHRAC1 gene (Gene ID 54108).
  • Forms a complex with DNA polymerase epsilon subunit POLE3 and binds naked DNA, which is then incorporated into chromatin, aided by the nucleosome remodeling activity of ISWI/SNF2H and ACF1. Does not enhance nucleosome sliding activity. Related references: PMID:14759371
  • Predominantly localized to the nucleus, consistent with its role in chromatin dynamics.
  • Widely expressed across human tissues, including heart, brain, placenta, lung, liver, skeletal muscle, kidney, and pancreas.
  • Annotated with post-translational modifications such as acetylation and phosphorylation, suggesting regulatory mechanisms.
  • Classified under keywords including DNA-binding, coiled-coil, nucleotidyltransferase, and phosphoprotein, highlighting its functional versatility.

Experimental Guidance and Technical Tips

  • For Western blot, consider starting with a dilution range of 1:500–1:2000 and optimize based on your specific sample type and detection system.
  • In ELISA, use the recombinant immunogen or purified CHRAC1 protein for coating to determine optimal antibody titration.
  • The antibody was raised against an N-terminal fragment (aa 1–131) and may detect both full-length and processed forms; validate in human cell lines or tissue lysates relevant to your experimental context.
  • Cross-reactivity is confirmed for human; if testing other species, perform sequence alignment and preliminary assays to assess reactivity.
