Biological Background: CHPT1 Function and Localization
- CHPT1 (also known as CPT1 or CPT) is a cholinephosphotransferase that catalyzes the final step of de novo phosphatidylcholine (PC) synthesis, transferring choline phosphate from CDP-choline to diacylglycerol (DAG).
- The protein is integral to membrane biogenesis, playing a central role in the formation and maintenance of vesicular membranes.
- CHPT1 localizes to the Golgi apparatus membrane.
- It is highly expressed in testis, colon, small intestine, heart, prostate, and spleen; detected at lower levels in several other tissues; and is overexpressed in cancerous breast epithelial cell lines.
- The enzyme requires magnesium or manganese as cofactors and contains at least one transmembrane helix.
- The CHPT1 gene undergoes alternative splicing, and the protein is subject to acetylation.
- A 3D structure of the protein has been solved, providing insights into its enzymatic mechanism.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to the N-terminal region (amino acids 1-100) of human CHPT1. This antibody may recognize full-length CHPT1 in denaturing western blot; validate in your sample system.
- For ELISA applications, consider using the immunogen peptide as a positive control to assess antibody binding efficiency.
- Optimize antibody dilution for WB and ELISA based on your experimental conditions. If recommended dilutions are not available, perform a titration curve.
- When preparing lysates, include appropriate detergents to solubilize membrane proteins, as CHPT1 is a Golgi membrane protein.
