Biological Background: Chondrolectin Function and Localization
- Chondrolectin (CHODL) is a transmembrane protein also known as MT75, encoded by the CHODL gene (C21orf68).
- It may function in nervous system development, potentially promoting neurite outgrowth and elongation, as well as motor axon growth and guidance.
- CHODL exhibits alternative promoter usage and alternative splicing, generating multiple isoforms; isoform 2 is predominantly expressed in B lymphocytes and mature thymocytes, while isoform 3 is specifically expressed in peripheral blood T lymphocytes.
- The protein localizes to the cytoplasm, membranes, and endoplasmic reticulum (ER) membrane.
- CHODL is a lectin family member containing a signal peptide, a transmembrane helix, and disulfide bonds; it is classified as a glycoprotein.
- Tissue expression is observed in spleen, testis, prostate, fetal liver, heart, skeletal muscle, and small intestine, with isoform-specific patterns in immune cells.
Experimental Guidance and Technical Tips
- The immunogen consists of a recombinant fusion protein spanning amino acids 110–210 of human CHODL. As a polyclonal antibody, it is expected to recognize multiple epitopes within this region.
- For Western blot (WB), a band around 30 kDa (consistent with the predicted molecular weight of 30–31 kDa) may be observed. Due to alternative splicing, additional isoforms may appear; validate in appropriate tissue or cell lysates.
- Given the cross-reactivity with mouse and rat, the antibody may be suitable for detecting orthologs in these species; however, sequence homology in the immunogen region should be checked.
- For ELISA, the recombinant protein fragment (or a peptide from the same region) could be used as a coating antigen. Titration and optimization are recommended.
