Biological Background: CHMP2B Function and Localization
- CHMP2B, also known as CHMP2.5 or Vps2-2, is a core component of the endosomal sorting complex required for transport III (ESCRT-III).
- It plays a critical role in multivesicular body (MVB) formation by assisting the invagination and scission of endosomal membranes to generate intraluminal vesicles (ILVs), which target cargo for lysosomal degradation.
- The ESCRT-III complex, including CHMP2B, functions in topologically equivalent membrane fission events such as cytokinesis and budding of enveloped viruses like HIV-1.
- CHMP2B localizes to the cytosol and late endosome membrane.
- It is widely expressed in human tissues with prominent expression in brain regions including cerebellum, cerebral cortex, medulla, and spinal cord, as well as in heart, skeletal muscle, spleen, kidney, liver, small intestine, pancreas, lung, placenta, and leukocytes.
- Post-translational modifications include acetylation and phosphorylation, and the protein is associated with coiled-coil structure.
- Mutations in CHMP2B are linked to amyotrophic lateral sclerosis (ALS) and other neurodegenerative disorders, classifying it as a disease-associated protein.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 114–213 of human CHMP2B, a region likely conserved across species (human, mouse, rat). This supports reliable detection across these species.
- For Western blotting (WB), a single band of approximately 24 kDa is expected in suitable cell or tissue lysates.
- For immunohistochemistry on paraffin-embedded sections (IHC-P), appropriate antigen retrieval and antibody dilution optimization may be required. Validate in the relevant sample system.
- For ELISA applications, the peptide-based immunogen design allows for sensitive detection of the target protein or peptide.
- Consider employing this monoclonal antibody as a detection reagent in ESCRT-III pathway studies, including membrane fission and endosomal sorting assays.
