Biological Background: CHMP2A Function and Localization
- CHMP2A (Charged multivesicular body protein 2a), also known as BC-2 or VPS2-1, is a core component of the endosomal sorting complex required for transport III (ESCRT-III).
- It plays a critical role in multivesicular body (MVB) formation, facilitating the sorting of endosomal cargo proteins into intraluminal vesicles destined for lysosomal degradation.
- The ESCRT-III complex, including CHMP2A, is essential for topologically equivalent membrane fission events such as the terminal stages of cytokinesis. Related references: PMID:21310966
- Together with SPAST, CHMP2A contributes to nuclear envelope sealing and mitotic spindle disassembly during late anaphase. Related references: PMID:26040712
- During nuclear envelope reformation, CHMP2A is recruited to the reforming nuclear envelope by LEMD2. Related references: PMID:28242692
- CHMP2A is also implicated in the budding of enveloped viruses, such as HIV-1, by participating in the ESCRT-mediated virus release pathway.
- Subcellularly, CHMP2A localizes to late endosome membranes and the nuclear envelope.
- Key post-translational modifications include acetylation and phosphorylation, and the protein features a coiled-coil domain.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein covering the full-length human CHMP2A (aa 1-222). Ensure specificity by validating with appropriate controls such as knockdown or knockout lysates if available.
- For Western blot (WB), the predicted molecular weight of CHMP2A is 25 kDa; consider using a suitable molecular weight marker and optimize blocking conditions to minimize background.
- For ELISA, the antibody can be used to detect recombinant CHMP2A or endogenous protein in cell lysates; include a standard curve for quantitative analysis.
- Cross-reactivity is reported for human; if using other species, verify sequence homology and perform a pilot experiment.
- Store and handle according to standard antibody protocols; shipped on ice bags to maintain stability.
