Biological Background: CHM Function and Localization

  • CHM, also known as Rab escort protein 1 (REP-1) or choroideremia protein, is a component of the Rab geranylgeranyltransferase (GGTase) complex that facilitates prenylation of Rab GTPases, essential for their membrane association and function.
  • It acts as a substrate-binding subunit, presenting unprenylated Rab proteins to the catalytic component B (RABGGTA/RABGGTB) and remaining bound after geranylgeranyl transfer before recycling the prenylated Rab to donor membranes.
  • A pre-formed CHM/Rab GGTase dimer (RGGT) complex can also directly bind and prenylate Rab proteins, proposed as the predominant pathway for Rab protein geranylgeranylation.
  • The gene CHM undergoes alternative splicing, and mutations are associated with choroideremia, a progressive retinal degeneration leading to vision loss.
  • CHM localizes predominantly to the cytoplasm and cytosol, consistent with its role in intracellular protein trafficking.
  • It belongs to the Rab escort protein family and contains motifs enabling GTPase activation.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 120–210 of human CHM, which is located within the central region of the protein; assess reactivity in relevant species and tissues.
  • For Western blot analysis, expect a band around 73 kDa; optimize antibody concentration and blocking conditions for your specific sample type.
  • ELISA application enables detection and quantification of CHM protein; consider pairing with a suitable capture antibody for sandwich ELISA setups.
