• CHM, also known as Rab escort protein 1 (REP-1) or choroideremia protein, is a component of the Rab geranylgeranyltransferase (GGTase) complex that facilitates prenylation of Rab GTPases, essential for their membrane association and function.
• It acts as a substrate-binding subunit, presenting unprenylated Rab proteins to the catalytic component B (RABGGTA/RABGGTB) and remaining bound after geranylgeranyl transfer before recycling the prenylated Rab to donor membranes.
• A pre-formed CHM/Rab GGTase dimer (RGGT) complex can also directly bind and prenylate Rab proteins, proposed as the predominant pathway for Rab protein geranylgeranylation.
