Biological Background: CHM (Rab escort protein 1) Function and Localization
- CHM encodes Rab proteins geranylgeranyltransferase component A 1, also known as Rab escort protein 1 (REP-1) or TCD protein, with a calculated molecular weight of approximately 73 kDa.
- It functions as the substrate-binding subunit of the Rab geranylgeranyltransferase (GGTase) complex, binding unprenylated Rab proteins and presenting them to the catalytic component B for geranylgeranylation.
- After the transfer reaction, CHM remains bound to the prenylated Rab and is thought to be regenerated by transferring the prenylated Rab back to the donor membrane, facilitating Rab protein membrane association.
- Alternatively, a pre-formed complex of CHM and the Rab GGTase dimer can directly bind and prenylate Rab proteins, which is proposed as the predominant pathway for Rab protein geranylgeranylation.
- Subcellularly, CHM localizes to the cytoplasm and cytosol, consistent with its role in Rab prenylation and trafficking.
- CHM is associated with choroideremia, an X-linked retinal degenerative disease, and is involved in sensory transduction and vision.
- Post-translationally, it undergoes alternative splicing, producing multiple isoforms, and is a reference proteome component with roles in GTPase activation.

- The immunogen corresponds to the N-terminal region (amino acids 1-100) of human CHM, which may recognize full-length CHM protein in denaturing Western blotting and native ELISA formats.
- For Western blotting, consider a starting dilution of 1:500-1:2000 and validate in human cell lines or tissues where CHM is expressed, such as retinal or lymphocyte samples.
- For ELISA, the antibody can be used as a capture or detection reagent; optimize coating conditions and antibody concentrations using recombinant CHM protein or cell lysates.
- Since CHM is a cytosolic protein, prepare samples using appropriate lysis buffers without detergents that may interfere with protein-protein interactions if studying complex formation.
