  • Chk2, encoded by the CHEK2 gene (also known as CDS1, RAD53, HuCds1), is a serine/threonine-protein kinase and tumor suppressor.
  • It phosphorylates numerous effectors at the consensus sequence [L-X-R-X-X-S/T], regulating cell cycle arrest, DNA repair, and apoptosis in response to DNA double-strand breaks. Related references: PMID:37943659
  • Chk2 inhibits CDC25 phosphatases to block CDK-cyclin activation, phosphorylates p53/TP53 at Ser-20 to stabilize it, and phosphorylates BRCA2 to promote homologous recombination repair.
