Biological Background: Chk1 Function and Localization
- Chk1 (Checkpoint kinase-1) is a serine/threonine-protein kinase encoded by the CHEK1 gene, functioning as a key cell cycle checkpoint kinase.

- Recognizes the substrate consensus sequence [R-X-X-S/T] and phosphorylates CDC25A, CDC25B, and CDC25C, leading to their inhibition via 14-3-3 binding or proteasomal degradation, thereby blocking cell cycle progression. - For Western blotting, a band around 54 kDa is expected. Thymus or testis lysates, which express high Chk1 levels, may serve as positive controls.
- When performing IHC-P or IF/ICC, consider optimization of fixation and permeabilization to enhance detection of nuclear and centrosomal Chk1.
- ELISA can be used for quantifying Chk1 in cell lysates or recombinant protein preparations; calibration against a standard curve in the specific matrix is advised.
