Biological Background: Chk1 Function and Localization
- Encoded by the CHEK1 gene, serine/threonine-protein kinase Chk1 undergoes alternative splicing to produce multiple isoforms, including an endogenous repressor isoform that antagonizes full-length Chk1 to promote the S to G2/M phase transition.

- Phosphorylates CDC25A, CDC25B and CDC25C, creating 14-3-3 binding sites or priming them for NEK11-mediated polyubiquitination and degradation, thereby inhibiting CDK-cyclin complexes and blocking cell cycle progression.
