Biological Background: Chk1 Function and Localization
- Chk1 (Checkpoint kinase 1) is a serine/threonine-protein kinase and a member of the protein kinase superfamily, with a conserved kinase domain and ATP-binding pocket.
- It is required for checkpoint-mediated cell cycle arrest and activation of DNA repair in response to DNA damage or unreplicated DNA, recognizing the substrate consensus sequence [R-X-X-S/T]. Related references: PMID:11535615 PMID:12399544 PMID:12446774
- Chk1 phosphorylates the CDC25 phosphatases (CDC25A, CDC25B, CDC25C), creating 14-3-3 binding sites and promoting CDC25A proteolysis, thereby inhibiting CDK-cyclin complexes and blocking cell cycle progression. Related references: PMID:12676583 PMID:12676925 PMID:12759351 PMID:9278511 PMID:19734889 PMID:20090422
- Phosphorylation of TP53 at multiple C-terminal sites by Chk1 promotes its acetylation and activation, leading to cell cycle arrest and suppression of proliferation. Related references: PMID:10673501 PMID:15659650 PMID:16511572
- Chk1 plays direct roles in DNA repair by phosphorylating RAD51 at Thr-309 (enhancing homologous recombination), FANCE (facilitating DNA cross-link repair), and regulating replication fork maintenance through PCNA. Related references: PMID:15665856 PMID:17296736 PMID:18451105
- Chk1 modulates chromatin through phosphorylation of TLK1 at Ser-743, preventing ASF1A phosphorylation and potentially enhancing chromatin assembly, and by phosphorylating histone H3.1 at Thr11 to regulate gene transcription. Related references: PMID:12660173 PMID:12955071
- An alternatively spliced isoform of CHEK1 acts as an endogenous repressor of full-length Chk1, promoting the S to G2/M phase transition through direct interaction and antagonism.
- Chk1 localizes to the nucleus, chromosomes, cytoplasm, and centrosomes. It is expressed ubiquitously with highest levels in thymus, testis, small intestine, and colon.
Experimental Guidance and Technical Tips
- For Western blotting, use reducing conditions and prepare lysates from cells or tissues known to express Chk1, such as thymus or testis. The predicted molecular weight is ~54 kDa; however, post-translational modifications may cause apparent shifts.
- The immunogen corresponds to amino acids 148-307 of human Chk1, which lies within the kinase domain. While this region is highly conserved in the canonical isoform, validation with appropriate controls is recommended to assess reactivity with potential alternative isoforms.
- This rabbit polyclonal antibody is validated for ELISA and WB. For ELISA applications, consider using recombinant Chk1 protein or fragment as a standard, and optimize antibody dilution and blocking conditions for your specific assay format.
- As a polyclonal antibody, lot-to-lot variability is possible. We recommend validating performance in your experimental system and including appropriate positive and negative controls.
