Biological Background: CHGA Function and Localization
- Chromogranin-A is a member of the granin family of neuroendocrine secretory proteins, also known as pituitary secretory protein I (SP-I).
- It strongly inhibits glucose-induced insulin release from the pancreas and acts as a non-competitive nicotinic cholinergic antagonist to inhibit catecholamine release from chromaffin cells and noradrenergic neurons. Related references: PMID:15326220
- Displays antibacterial activity against Gram-positive (S.aureus, M.luteus) and Gram-negative (E.coli, P.aeruginosa) bacteria, and acts as a potent free radical scavenger. Related references: PMID:15723172, PMID:24723458
- Can induce mast cell migration, degranulation, and production of cytokines and chemokines. Related references: PMID:21214543
- May play a role in the regulation of cardiac function and blood pressure. Related references: PMID:18541522
- Regulates granule biogenesis in endocrine cells by up-regulating SERPINE2 transcription via a cAMP-PKA-SP1 pathway, promoting granule formation.
- Secreted and found in cytoplasmic secretory vesicles, including neuronal dense core vesicles; detected in cerebrospinal fluid and urine.
- Undergoes amidation, sulfation, phosphorylation, glycosylation, and proteolytic cleavage, with disulfide bonds and calcium-binding properties.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 230–380 of human CHGA; this central region may be conserved across variants; verify reactivity in your experimental system.
- For Western blot, a band near 51 kDa is expected, though endogenous proteolytic processing may yield additional specific bands; adjust sample preparation and antibody dilution accordingly.
- In ELISA, consider pairing with a detection antibody recognizing a distinct epitope or use recombinant full-length CHGA as a standard for quantitative assays.
- Validate performance in neuroendocrine cell lines (e.g., PC12, SH-SY5Y) or relevant tissue lysates, as CHGA expression is tissue-specific.
