Biological Background: Chemerin Function and Localization
- Chemerin, also known as retinoic acid receptor responder protein 2 (RARRES2), is an adipocyte-secreted protein (adipokine) that regulates adipogenesis, metabolism, and inflammation.
- It acts as a ligand for the chemokine-like receptors CMKLR1 and CMKLR2, and can also bind to CCRL2 with lower affinity.
- Chemerin positively regulates adipocyte differentiation and modulates the expression of genes involved in lipid and glucose metabolism, potentially playing a role in angiogenesis in adipose tissue.
- It exhibits both pro- and anti-inflammatory properties depending on proteolytic cleavage; it can promote secretion of pro-inflammatory adipokines or exert anti-inflammatory effects by inhibiting VCAM1 expression and monocyte adhesion via AKT1/NOS3 signaling.
- As a chemotactic factor, Chemerin attracts leukocyte populations expressing CMKLR1, including dendritic cells, macrophages, and natural killer cells.
- In the skin, Chemerin displays an antimicrobial function.
- The protein contains a signal peptide, is secreted, and features a disulfide bond in its mature form (UniProt keyword: Disulfide bond).
- Chemerin is abundantly expressed in the liver and adipose tissue (including visceral, epididymal, and brown adipose tissue) and is detected at the protein level in differentiated adipocytes.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 21–162 of mouse Chemerin, which encompasses the region after the predicted signal peptide cleavage site; the antibody may recognize both full-length and cleaved forms.
- For Western blot, consider optimizing conditions with various tissue lysates where Chemerin is expressed (e.g., liver, adipose tissue).
- For ELISA, the polyclonal nature may provide robust detection; validate assay conditions with recombinant protein standards.
- Because cross-reactivity is reported for rat, this antibody can be used in comparative studies.
- Note that Chemerin undergoes proteolytic processing, so multiple bands may be observed in WB.
