- Serine/threonine-protein kinase Chk2 (also known as CHK2, Cds1, Rad53) is a critical checkpoint kinase involved in the DNA damage response.
- Functions include cell cycle arrest at G1/S and G2/M via phosphorylation of CDC25 phosphatases, leading to inhibition of CDK-cyclin complexes and blocking cell cycle progression.
- Promotes DNA repair by phosphorylating BRCA2, enhancing RAD51-mediated homologous recombination; also transcriptionally activates DNA repair genes (e.g., BRCA2) via FOXM1.
- Regulates apoptosis through phosphorylation of p53/TP53, MDM4, and PML, contributing to stabilization and activation of pro-apoptotic pathways.
- Acts as a tumor suppressor; may have a DNA damage-independent role in mitotic spindle assembly by phosphorylating BRCA1.
