Biological Background: CHD9 Function and Localization
- Known as ATP-dependent chromatin remodeler CHD9, also called Chromatin-related mesenchymal modulator (CReMM), CHD-9, or Kismet homolog 2 (KISH2).
- Functions as a transcriptional coactivator for PPARA and likely other nuclear receptors, promoting gene transcription.
- Possesses DNA-dependent ATPase activity and preferentially binds to A/T-rich DNA regions.
- Associates with A/T-rich regulatory regions in promoters of genes involved in osteogenic differentiation of progenitor cells (By similarity).
- Subcellular localization includes both cytoplasm and nucleus.
- Widely expressed at low levels, with restricted expression in bone marrow to osteoprogenitor cells adjacent to mature osteoblasts.
- Contains chromodomains, a helicase-like ATPase domain, and undergoes alternative splicing; post-translationally modified by acetylation, phosphorylation, and ubiquitination.
Experimental Guidance and Technical Tips
- For Western blotting, note the large protein size (predicted ~326 kDa). Use low-percentage SDS-PAGE gels (e.g., 6% or gradient) and wet transfer with extended transfer time to ensure efficient transfer.
- The immunogen corresponds to amino acids 85–190, an N-terminal region; this may allow detection of full-length CHD9 and possibly N-terminal-containing isoforms.
- In ELISA, consider direct coating of recombinant CHD9 protein or peptide fragment for assay optimization.
- Verify antibody reactivity in your target species (human and mouse) with appropriate positive and negative controls.
