Biological Background: CHCHD4 Function and Localization
- CHCHD4, also known as MIA40 or TIMM40, is a mitochondrial intermembrane space import and assembly protein essential for the biogenesis of respiratory chain complexes. Related references: PMID:26004228
- It functions as a redox-sensitive chaperone, catalyzing the formation of disulfide bonds in substrate proteins such as COA7, COX17, COX19, MICU1, and TIMCC. Related references: PMID:16185709, PMID:19182799, PMID:21059946
- CHCHD4 is required for the import and folding of small cysteine-containing proteins in the mitochondrial intermembrane space, including COA7. Related references: PMID:30885959
- The import mechanism involves a disulfide relay: reduced precursor proteins are oxidized via a transient intermolecular disulfide bridge with CHCHD4, promoting folding; CHCHD4 is then reoxidized by GFER/ERV1. Related references: PMID:16185709, PMID:19182799, PMID:21059946, PMID:23186364
- Subcellularly, CHCHD4 localizes to the mitochondrial intermembrane space.
- It is ubiquitously expressed across all tested human tissues.
- Alternative splicing yields multiple isoforms, and the protein contains characteristic coiled-coil-helix-coiled-coil-helix domains.
- Post-translational modifications include disulfide bonds and a redox-active center crucial for its oxidoreductase activity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1-105), so this antibody may recognize all major isoforms if the epitope is conserved; validate specificity in your sample system.
- For Western blotting, the predicted molecular weight is approximately 16 kDa; consider using a suitable gel percentage and membrane to resolve small proteins.
- For immunohistochemistry (paraffin), optimize antigen retrieval and primary antibody concentration using appropriate positive and negative tissue controls.
- In ELISA applications, validate detection of the target protein by comparing signals from recombinant protein standards and endogenous samples.
- Cross-reactivity with rat is indicated; when working with other species, verify reactivity experimentally.
