Biological Background: CHCHD3/MIC19 Function and Localization

  • CHCHD3 (Coiled-coil-helix-coiled-coil-helix domain-containing protein 3), also known as MIC19, MINOS3, MICOS19, or PPP1R22, is a component of the mitochondrial contact site and cristae organizing system (MICOS) complex. It is a nuclear-encoded protein with a predicted molecular weight of ~26 kDa (UniProt: 26152 Da) and shares homology with other CHCHD family members.
  • As a key subunit of the MICOS complex, CHCHD3 localizes to the mitochondrial inner membrane and is essential for maintaining crista junction architecture and inner membrane organization. It also facilitates contact site formation between inner and outer mitochondrial membranes (PubMed:25781180, PubMed:32567732, PubMed:33130824).

  • Beyond its structural role, CHCHD3 has been implicated in transcriptional regulation. It can localize to the nucleus and act as a repressor by binding to the BAG1 promoter, thereby downregulating BAG1 expression (PubMed:22567091).

  • CHCHD3 is subject to multiple post-translational modifications, including acetylation, phosphorylation, and myristoylation, which may regulate its dual localization and functions. It contains conserved disulfide bonds critical for its structural integrity.
  • In human tissues, CHCHD3 is widely expressed at low levels but shows enriched expression in heart and skeletal muscle. Notably, its expression is upregulated in primary lung cancers compared to normal lung tissue, suggesting a potential role in tumorigenesis.

Experimental Guidance and Technical Tips

  • For Western blot analysis, a band corresponding to ~26 kDa is predicted; however, we recommend validating the observed molecular weight in your sample system, as post-translational modifications may affect migration.
  • When performing immunofluorescence (IF/ICC), expect mitochondrial localization. Co-staining with a mitochondrial marker is advisable to confirm targeting specificity.
  • For immunoprecipitation (IP) experiments, optimize lysis conditions and include an appropriate control antibody to ensure specific capture.
  • As a rabbit polyclonal antibody raised against the N-terminal fragment (amino acids 1–227), it may detect full-length CHCHD3 and potentially certain isoforms. Specificity should be validated in the relevant biological model.
