Biological Background: CHAC1 Function and Localization
- CHAC1 (Glutathione-specific gamma-glutamylcyclotransferase 1), also known as Botch or cation transport regulator-like protein 1, is a 24.4 kDa protein encoded by the CHAC1 gene (Gene ID: 79094).
- It acts as a glutathione-specific gamma-glutamylcyclotransferase, cleaving glutathione into 5-oxo-L-proline and Cys-Gly, with specificity for glutathione over other gamma-glutamyl peptides. Related references: PMID:27913623
- CHAC1 promotes apoptosis by depleting glutathione and functions as a pro-apoptotic component of the unfolded protein response (UPR) pathway, mediating the effects of the ATF4-ATF3-DDIT3/CHOP cascade. Related references: PMID:19109178
- It negatively regulates Notch signaling during embryonic neurogenesis by inhibiting Notch cleavage by furin, maintaining Notch in an immature inactive form, thereby promoting neurogenesis. Related references: PMID:22445366
- Subcellularly, CHAC1 localizes to the cytoplasm, cytosol, and the Golgi apparatus, particularly the trans-Golgi network.
- Keywords: alternative splicing, apoptosis, lyase, neurogenesis, Notch signaling pathway, unfolded protein response.
Experimental Guidance and Technical Tips
- The antibody was raised against a recombinant protein spanning amino acids 1-222 (full-length CHAC1, NP_077016.3), which may recognize multiple epitopes and is expected to detect endogenous CHAC1.
- Consider using a denaturing SDS-PAGE gel for western blotting, where the predicted molecular weight of CHAC1 is ~24 kDa; validate the band size in your sample system.
- Appropriate positive controls include cell lines or tissues known to express CHAC1 under unfolded protein response conditions (e.g., ER stress induction); include a loading control.
- For immunofluorescence and immunohistochemistry, optimize fixation and permeabilization conditions, and consider antigen retrieval for paraffin-embedded sections.
- Cross-reactivity is reported for human, mouse, and rat; however, pre-testing in your model system is recommended to verify reactivity and minimize non-specific binding.
- ELISA applications may require optimization of coating conditions and antibody dilution; a direct or indirect ELISA format can be explored depending on assay design.
