Biological Background: CGRP (Calcitonin Gene-Related Peptide) Function and Localization
- The CALCA gene undergoes alternative splicing to produce two peptide hormones: calcitonin and calcitonin gene-related peptide (CGRP, also known as CGRP1, CGRP-alpha, CGRP-I). This antibody specifically recognizes the CGRP isoform.
- Calcitonin, encoded by the same gene, is a 32-amino acid peptide hormone that rapidly reduces blood calcium and phosphate levels by promoting their deposition in bones. However, the antibody was raised against the CGRP-specific sequence.

- Katacalcin, another cleavage product from the precursor, also exhibits calcium-lowering activity.

Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning the CGRP pro-region (aa 49–128). For Western blot, bands at ~13 kDa and/or ~15 kDa may be observed, corresponding to precursor forms; mature CGRP (~3.8 kDa) may run at the dye front.
- Consider using low-percentage tricine gels or specialized electrophoresis systems to resolve small peptides if detection of mature CGRP is desired.
- Blocking with BSA rather than milk may reduce background, as CGRP is a small, highly modified peptide.
- For immunohistochemistry and immunofluorescence, perform antigen retrieval suited to the tissue and validate staining in known CGRP-positive tissues (e.g., sensory neurons, cardiovascular tissue).
- Cross-reactivity is reported with mouse and rat; however, endogenous expression levels vary among tissues and disease states, so optimization of sample preparation is advised.
