Biological Background: CGPR Function and Localization
- Calcitonin gene-related peptide 2 (CGPR), also known as beta-CGRP or CGRP-II, is encoded by the CALCB (CALC2) gene and belongs to the calcitonin peptide family.
- It functions as a potent vasodilator, inducing relaxation of various vessels including coronary, cerebral and systemic vasculature through the CALCRL-RAMP1 receptor complex. Related references: PMID:1318039 PMID:9620797
- Abundant expression in the central nervous system suggests a role as a neurotransmitter or neuromodulator. Related references: PMID:3492492
- Subcellular location: Secreted into the extracellular space.
- Tissue specificity: Predominantly expressed in spinal cord, pituitary, and thalamus.
- Post-translational modifications include amidation, disulfide bond formation, and cleavage on pair of basic residues, essential for its biological activity.
- Mature molecular weight is approximately 13.7 kDa (UniProt; product listed at 13 kDa).
Experimental Guidance and Technical Tips
- The immunogen corresponds to residues 20-120 of the human CGPR precursor (NP_000719.1), which includes the propeptide and mature peptide regions. The antibody may detect both precursor and fully processed forms.
- For Western blot, consider using samples enriched for secreted peptides, such as serum, plasma, or conditioned medium, or tissues with high CGPR expression (e.g., spinal cord). Ensure protein extraction and handling preserve low-molecular-weight peptides.
- Cross-reactivity with mouse allows testing in mouse tissues; however, validate the antibody’s performance in your specific model.
- For ELISA, the antibody can be used as capture or detection reagent after optimization with appropriate peptide standards.
- Note that the mature CGPR peptide is only ~13 kDa, so use high-percentage gels (e.g., 15%) and appropriate molecular weight markers for resolution.
