Biological Background: CGGBP1 Function and Localization
- CGGBP1 (CGG triplet repeat-binding protein 1) is a DNA-binding protein also known as 20 kDa CGG-binding protein (p20-CGGBP). It belongs to the reference proteome and has been identified by proteomics and direct protein sequencing.
- CGGBP1 specifically binds to nonmethylated 5'-d(CGG)(n)-3' trinucleotide repeats in the FMR1 promoter, and may play a role in regulating FMR1 promoter activity, implicating it in transcription regulation.
- Localized predominantly in the nucleus, consistent with its DNA-binding function.
- Ubiquitously expressed across tissues, with high expression in placenta, thymus, lymph nodes, cerebellum, and cerebral cortex; low expression in other brain regions.
- Post-translational modifications include phosphorylation (phosphoprotein keyword), which may regulate its activity.
- The calculated molecular weight is approximately 18.8 kDa, consistent with the observed 19 kDa band.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1–167) of human CGGBP1; consider using full-length or truncated recombinant protein as a positive control.
- For Western blot, a band at approximately 19 kDa is expected; validate detection in your specific sample system using appropriate loading controls.
- For ELISA, the antibody may be used for direct or indirect detection; perform titration to determine optimal working concentration.
- Given nuclear localization, nuclear extraction protocols may enrich the target protein and improve signal.
