Biological Background: cGAS Function and Localization
- cGAS (cyclic GMP-AMP synthase), also known as MB21D1 or C6orf150, is encoded by the CGAS gene. Alternative splicing generates multiple isoforms.
- It functions as a nucleotidyltransferase that catalyzes the synthesis of the cyclic dinucleotide 2'3'-cGAMP from ATP and GTP. Related references: PMID:21478870, PMID:23258413, PMID:23707061, PMID:28214358, PMID:28363908
- cGAS is a pivotal cytosolic DNA sensor that directly binds double-stranded DNA (dsDNA) of sufficient length and curvature, leading to liquid-phase condensation and enzymatic activation. Related references: PMID:28314590, PMID:28363908, PMID:29976794, PMID:30007416
- Upon activation, cGAS-produced 2'3'-cGAMP binds the adaptor STING1, triggering a signaling cascade that induces type-I interferons and other immune mediators. Related references: PMID:28314590, PMID:28363908, PMID:29976794
- cGAS detects dsDNA from a wide range of pathogens including DNA viruses, retroviruses (e.g., HIV-2), and bacteria (e.g., M. tuberculosis), making it a broad innate immune sentinel. Related references: PMID:28363908, PMID:35613581, PMID:23929945, PMID:24269171, PMID:30270045, PMID:26048138
- Under cellular stress (mitochondrial damage, senescence, DNA damage), self-DNA leaks into the cytoplasm or accumulates as micronuclei, aberrantly activating cGAS and driving sterile inflammation. Related references: PMID:28738408, PMID:28759889, PMID:31299200
- cGAS resides predominantly in the cytoplasm but is also found in the nucleus, where its binding to nucleosomes on chromatin inhibits its activity to prevent autoimmune activation. Related references: PMID:31299200, PMID:32911482, PMID:32912999
- Beyond immunity, cGAS modulates DNA damage repair by interacting with PARP1 to suppress homologous recombination and senses translational stress by associating with collided ribosomes. Related references: PMID:30356214, PMID:31544964, PMID:34111399
- Tissue expression is reported in monocytic cell line THP1; post-translational modifications (acetylation, phosphorylation, ubiquitination, palmitoylation) dynamically regulate cGAS localization and activity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a peptide within the C-terminal region (aa 430–522) of human cGAS. Consider verifying recognition of all splice variants if multiple isoforms are relevant.
- For Western blot (WB), a band of approximately 59 kDa is expected; use appropriate positive controls (e.g., THP1 cell lysate). Validate antibody specificity using cGAS knockout or knockdown samples.
- For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), optimized antigen retrieval and permeabilization conditions may be required. Include appropriate controls to confirm nuclear/cytoplasmic localization.
- In ELISA applications, ensure the coating buffer and detection conditions are compatible with the rabbit monoclonal format.
- Cross-reactivity has been tested only against human samples; reactivity with other species is not characterized and should be experimentally determined.
