Biological Background: CASP8 and FADD-like apoptosis regulator Function and Localization

• Acts as an apoptosis regulator protein that may function as a crucial link between cell survival and cell death pathways in mammalian cells. Lacks enzymatic caspase activity. Related references: PMID:31046799
• Functions as an inhibitor of TNFRSF6 (Fas)-mediated apoptosis. A proteolytic fragment (p43) is retained in the death-inducing signaling complex (DISC), blocking further recruitment and processing of caspase-8.

Experimental Guidance and Technical Tips

• For Western blot, a band around 55 kDa is expected for full-length cFLIP. Additional bands corresponding to isoforms or cleavage products (e.g., p43 fragment) may be observed; validate in your sample system.
• Since the immunogen covers amino acids 90-240, which is a region shared by some isoforms, the antibody may detect multiple isoforms. Specific isoform assignment should be confirmed with appropriate controls.
• For ELISA, appropriate coating antigen and antibody dilution should be empirically determined. Pairing with a detection antibody targeting a non-overlapping epitope may improve assay specificity.
