Biological Background: CASP8 and FADD-like apoptosis regulator Function and Localization
- Acts as an apoptosis regulator protein that may function as a crucial link between cell survival and cell death pathways in mammalian cells. Lacks enzymatic caspase activity. Related references: PMID:31046799
- Functions as an inhibitor of TNFRSF6 (Fas)-mediated apoptosis. A proteolytic fragment (p43) is retained in the death-inducing signaling complex (DISC), blocking further recruitment and processing of caspase-8.
- Full-length and shorter isoforms have been shown either to induce apoptosis or to reduce TNFRSF-triggered apoptosis, depending on the isoform.
- Widely expressed, with higher expression in skeletal muscle, pancreas, heart, kidney, placenta, and peripheral blood leukocytes. Isoform 8 is predominantly expressed in testis and skeletal muscle.

Experimental Guidance and Technical Tips
- For Western blot, a band around 55 kDa is expected for full-length cFLIP. Additional bands corresponding to isoforms or cleavage products (e.g., p43 fragment) may be observed; validate in your sample system.
- Since the immunogen covers amino acids 90-240, which is a region shared by some isoforms, the antibody may detect multiple isoforms. Specific isoform assignment should be confirmed with appropriate controls.
- For ELISA, appropriate coating antigen and antibody dilution should be empirically determined. Pairing with a detection antibody targeting a non-overlapping epitope may improve assay specificity.
- Perform initial experiments using positive control lysates from tissues with high cFLIP expression (e.g., skeletal muscle, pancreas, heart) or relevant cell lines.
