Biological Background: CETP Function and Localization
- Cholesteryl ester transfer protein (CETP), also known as lipid transfer protein I, facilitates the transfer of neutral lipids (cholesteryl ester and triglyceride) between lipoprotein particles. It enables the net movement of cholesteryl ester from high-density lipoproteins (HDL) to very low-density lipoproteins (VLDL) and the equimolar transport of triglyceride from VLDL to HDL. - CETP plays a central role in reverse cholesterol transport, removing excess cholesterol from peripheral tissues and returning it to the liver for elimination. Related references: PMID:17237796
- The protein is primarily expressed by the liver and secreted into the plasma, consistent with its role in systemic lipid metabolism.
- Subcellularly, CETP is a secreted protein that functions extracellularly in the bloodstream.
- UniProt keywords highlight its involvement in cholesterol metabolism, lipid transport, steroid metabolism, and atherosclerosis; it is also a glycoprotein with a disulfide bond.
- Alternative splicing of the CETP gene generates multiple isoforms, which may contribute to functional diversity.
Experimental Guidance and Technical Tips
- The immunogen used spans amino acids 300–493, a region that may offer specificity among CETP isoforms. Validate performance in your target sample type (e.g., human liver lysate or plasma).
- For Western blot, the predicted molecular weight is ~55 kDa; use appropriate loading controls and confirm band patterns in relevant biological replicates.
- In IHC-P, consider antigen retrieval methods and appropriate tissue fixation conditions, as CETP expression is primarily hepatic.
- Cross-reactivity has been shown for human; if testing other species, verify sequence homology and perform validation experiments.
