Biological Background: CES2 Function and Localization
- CES2, also known as cocaine esterase, carboxylesterase 2 (CE-2), and methylumbelliferyl-acetate deacetylase 2, is encoded by the CES2 gene (ICE). It belongs to the serine esterase family and is involved in the detoxification of xenobiotics and activation of ester and amide prodrugs. Related references: PMID:9169443
- It demonstrates high catalytic efficiency for hydrolyzing cocaine, 4-methylumbelliferyl acetate, heroin, and 6-monoacetylmorphine. Related references: PMID:9169443
- CES2 hydrolyzes aspirin and substrates with large alcohol groups and small acyl groups. It also plays a role in lipid metabolism, hydrolyzing endogenous lipids like triacylglycerol and converting monoacylglycerides to free fatty acids and glycerol. Related references: PMID:28677105
- The enzyme hydrolyzes 2-arachidonoylglycerol and prostaglandins, indicating involvement in endocannabinoid and prostaglandin metabolism. Related references: PMID:21049984
- Subcellularly, CES2 localizes to the endoplasmic reticulum lumen, consistent with its role in intracellular ester hydrolysis.
- Expression is preferential in the intestine, with highest levels in the small intestine, moderate in the liver, and lower in the colon and rectum.
- Post-translational modifications include signal peptide cleavage, N-glycosylation, and formation of pyrrolidone carboxylic acid at the N-terminus. Alternative initiation and splicing generate multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen targets a C-terminal region (amino acids 310–410) of human CES2; the antibody may recognize full-length protein as well as certain isoforms or cleavage products. Validate in your sample system.
- Predicted molecular weight by Western blot is ~62 kDa. Use appropriate reducing conditions and verify band specificity with positive and negative controls.
- For IF/ICC, consider optimizing fixation and permeabilization. Endoplasmic reticulum localization can guide expected staining patterns.
- Reported cross-reactivity with human and mouse; sequence homology suggests potential reactivity with other species—verify experimentally if needed.
