Biological Background: CES1 Function and Localization
- CES1 (liver carboxylesterase 1) is a serine esterase involved in the detoxification of xenobiotics and activation of ester and amide prodrugs. It hydrolyzes aromatic and aliphatic esters but has no catalytic activity toward amides or fatty acyl-CoA esters. Related references: PMID:18762277 PMID:7980644 PMID:9169443
- Hydrolyzes the methyl ester group of cocaine to form benzoylecgonine and catalyzes the transesterification of cocaine to form cocaethylene. Related references: PMID:7980644
- Possesses fatty acid ethyl ester synthase activity, catalyzing the ethyl esterification of oleic acid to ethyloleate. Related references: PMID:7980644
- Converts monoacylglycerides to free fatty acids and glycerol, and hydrolyzes 2-arachidonoylglycerol and prostaglandins. Related references: PMID:21049984
- Plays a key role in reverse cholesterol transport by hydrolyzing cholesteryl esters to free cholesterol, facilitating efflux from macrophages and release in the liver. Related references: PMID:11015575 PMID:16024911 PMID:16971496 PMID:18599737 PMID:18762277
- Expressed predominantly in liver, with lower levels in heart and lung, and detected in macrophages. Related references: PMID:10562416 PMID:11015575 PMID:18762277 PMID:21049984
- Localized to the endoplasmic reticulum lumen, cytoplasm, and lipid droplets.
- Subject to alternative splicing, glycosylation, and phosphorylation; contains a signal peptide and disulfide bonds.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the N-terminal region (aa 1–100) of human CES1, which may confer cross-reactivity to mouse and rat due to sequence conservation.
- For Western blot, the predicted molecular weight is ~63 kDa; consider using appropriate loading controls and optimizing antibody dilution based on sample type.
- In immunofluorescence (IF-P) experiments, validate fixation and permeabilization protocols to preserve antigenicity and subcellular localization.
- ELISA performance should be verified with recombinant CES1 protein or native samples; pairing with a detection antibody may require empirical optimization.
