Biological Background: CES1 Function and Localization
- CES1 (liver carboxylesterase 1) is a serine esterase also known by multiple aliases including acyl-coenzyme A:cholesterol acyltransferase (ACAT), cholesteryl ester hydrolase (CEH), triacylglycerol hydrolase (TGH), and cocaine carboxylesterase, reflecting its diverse substrate specificity and roles in lipid and xenobiotic metabolism.
- The enzyme is centrally involved in the detoxification of xenobiotics and the activation of ester and amide prodrugs. It hydrolyzes a broad range of aromatic and aliphatic esters, but shows no catalytic activity toward amides or fatty acyl-CoA esters. Related references: PMID:18762277, PMID:7980644, PMID:9169443
- CES1 catalyzes the hydrolysis of cocaine’s methyl ester group to benzoylecgonine and can also mediate the transesterification of cocaine to cocaethylene in the presence of ethanol. Related references: PMID:7980644
- In lipid metabolism, CES1 displays fatty acid ethyl ester synthase activity, converts monoacylglycerides to free fatty acids and glycerol, and hydrolyzes 2‑arachidonoylglycerol and prostaglandins. Related references: PMID:7980644, PMID:21049984
- The enzyme hydrolyzes intracellular cholesteryl esters, promoting reverse cholesterol transport (RCT) by facilitating free cholesterol efflux from macrophages and subsequent hepatic release of cholesterol for bile acid synthesis or direct biliary secretion. Related references: PMID:11015575, PMID:16024911, PMID:16971496, PMID:18599737, PMID:18762277
- Subcellularly, CES1 localizes primarily to the endoplasmic reticulum lumen, cytoplasm, and lipid droplets.
- Expression of CES1 is highest in the liver, with lower levels detected in heart and lung, and it is also present in macrophages. Related references: PMID:10562416, PMID:11015575, PMID:18762277, PMID:21049984
- Post‑translational modifications include N‑linked glycosylation and phosphorylation; the protein contains a signal peptide, intramolecular disulfide bonds, and undergoes alternative splicing to generate multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen comprises the C‑terminal segment (aa 287–566) of human CES1, which includes part of the catalytic domain; this design may generate antibodies capable of recognizing several CES1 isoforms.
- For western blotting under reducing conditions, expect a band near 63 kDa; endogenous expression is highest in liver and macrophage samples, so these should be considered as positive controls.
- For ELISA applications, using the recombinant immunogen or purified native protein as a standard can help validate assay sensitivity and specificity.
- Cross‑reactivity with mouse CES1 is stated; however, validation in the intended sample matrix and species is recommended to confirm performance.
