Biological Background: Ceruloplasmin Function and Localization
- Ceruloplasmin is a multifunctional blue copper-binding glycoprotein containing 6-7 copper atoms per molecule.
- It exhibits ferroxidase activity, oxidizing Fe(2+) to Fe(3+) without releasing radical oxygen species, which is crucial for iron transport across cell membranes. Related references: PMID:16150804
- The enzyme also catalyzes Cu(+) oxidation and promotes the oxidation of biogenic amines like norepinephrine and serotonin. Related references: PMID:14623105 PMID:4643313 PMID:5912351
- It provides Cu(2+) ions for the ascorbate-mediated deaminase degradation of heparan sulfate chains of GPC1 (By similarity).
- Ceruloplasmin possesses glutathione peroxidase-like activity, removing hydrogen peroxide and lipid hydroperoxide in the presence of thiols. Related references: PMID:10481051
- It acts as an inhibitor of the peroxidase activity of MPO (By similarity).
- Additionally, it shows NO-oxidase and NO2 synthase activities that help regulate endocrine NO homeostasis. Related references: PMID:16906150
- Ceruloplasmin is primarily expressed by the liver and secreted into plasma; it is a secreted glycoprotein with multiple disulfide bonds, repeated domains, and phosphorylation sites.
Experimental Guidance and Technical Tips
- As a rabbit polyclonal antibody, this product recognizes multiple epitopes; consider this advantage for detecting the target in various sample conditions.
- For immunofluorescence (IF-P) experiments, optimize fixation and permeabilization protocols suitable for secreted proteins.
- The immunogen corresponds to amino acids 680–790, a C-terminal region; validate detection of full-length ceruloplasmin in your ELISA setup.
- Cross-reactivity with mouse and rat suggests conserved epitopes; verify performance in the relevant species and tissue context.
