Biological Background: Ceruloplasmin Function and Localization

  • Ceruloplasmin, also known as ferroxidase or cuproxidase, is a secreted copper-binding glycoprotein encoded by the CP gene.
  • It exhibits ferroxidase activity, oxidizing Fe2+ to Fe3+ without generating radical oxygen species, which is critical for iron transport across membranes.   • The immunogen covers amino acids 366–572 of human ceruloplasmin; therefore, this antibody may detect both full-length and potential splice variants or fragments containing this region.
  • For Western blotting (WB), the predicted molecular weight of ceruloplasmin is approximately 122 kDa; consider using an appropriate gel percentage and optimized transfer conditions for high-molecular-weight proteins.
  • For immunohistochemistry (IHC-P) and immunofluorescence (IF-P), antigen retrieval and appropriate tissue preparation are recommended. Validate staining patterns in known positive and negative tissues.
  • In ELISA, the antibody can be used as either capture or detection reagent. Optimal pairing with a complementary anti-CP antibody should be empirically determined.
  • Due to sequence homology, cross-reactivity with mouse and rat ceruloplasmin is expected. Further validation in the relevant species model is advised.
