Biological Background: Ceramide Synthase 5 Function and Localization
- CERS5 (Ceramide synthase 5) is a multi-pass membrane protein of the endoplasmic reticulum, belonging to the Lass family of ceramide synthases.
- It catalyzes the N-acylation of sphingoid bases using fatty acyl-CoAs, with high selectivity for palmitoyl-CoA (C16:0-CoA), a key step in de novo ceramide synthesis. Related references: PMID:16951403, PMID:18541923, PMID:22144673
- CERS5 can also use various sphingoid bases as substrates, including sphinganine, sphingosine, phytosphingosine, and sphinga-4E,14Z-dienine, supporting both de novo and salvage pathways of ceramide production. Related references: PMID:31916624
- The enzyme contributes to lipid homeostasis in pulmonary epithelia, playing a role in surfactant production.
- Post-translational modifications include N-linked glycosylation and phosphorylation sites.
- CERS5 is encoded by the CERS5 gene (previously LASS5) and potentially generates multiple isoforms through alternative splicing.
Experimental Guidance and Technical Tips
- For Western blotting, optimize the primary antibody dilution using a concentration gradient, and validate in a relevant cell line or tissue expressing endogenous CERS5.
- A blocking peptide corresponding to the immunogen sequence can serve as a specificity control in both WB and ELISA.
- In ELISA, use the synthetic peptide antigen as a positive control to confirm antibody reactivity.
- Consider testing cross-reactivity in mouse and rat samples, as indicated, by comparing signal strengths and potentially performing species-specific blocking.
