Biological Background: CEP55 Function and Localization

  • CEP55 (Centrosomal protein of 55 kDa), also designated URCC6 (Up-regulated in colon cancer 6), is a coiled‑coil protein encoded by the CEP55 gene (C10orf3, URCC6). Alternative splicing yields multiple isoforms.
  • Plays an indispensable role in mitotic exit and cytokinesis by recruiting PDCD6IP and TSG101 to the midbody; not required for microtubule nucleation.
    Related references: PMID:16198290 PMID:17853893
  • Required for proper brain and kidney development.
    Related references: PMID:28264986
  • Subcellularly localizes to the cytoplasm, centrosome, cleavage furrow, and prominently to the midbody and midbody ring during cytokinesis.
  • Subject to phosphorylation and classified as a phosphoprotein.
  • Widely expressed in proliferative tissues, with highest levels in testis and cancer cell lines; also detected in embryonic brain, fetal brain ganglionic eminence, kidney tubules, adult brain, cerebellum, and intestine (at protein level).

Experimental Guidance and Technical Tips

  • The immunogen spans the N‑terminal 1–260 amino acids of human CEP55, covering a region that may be well‑conserved across species; validate cross‑reactivity in your sample system before quantitative assays.
  • For western blotting, consider using whole‑cell lysates from rapidly dividing cells (e.g., HeLa, HEK293) or testis tissue to enrich the target; the predicted molecular weight is ~54 kDa, but verify the band pattern using appropriate controls.
  • In immunofluorescence (IF/ICC), staining of mitotic cells can highlight midbody localization; optimize fixation and permeabilization conditions to preserve the midbody structure.
  • For ELISA, titrate the antibody and confirm specificity against recombinant protein or peptide; note that polyclonal lots may exhibit slight variation in affinity profiles.
